This position is part of a DFG-funded project on visual proteomics of Mycoplasma pneumoniae using advanced photocrosslinking mass spectrometry (crosslinking MS). The project aims to map protein-protein interactions in situ, interpret these interactions in the context of cellular structure, and explore their relevance for cellular mechanisms including those affected by antibiotics and other protein-targeting small molecules.

• Perform in-cell photocrosslinking using diazirine-based reagents to stabilise native protein interactions.
• Carry out large-scale sample preparation for proteome-wide crosslinking MS, including multidimensional fractionation (SEC, SCX, hSAX, RP).
• Assist with LC-MS/MS analyses using Orbitrap and Astral instrumentation in collaboration with MS specialists in the team.
• Contribute to analysis and interpretation of protein-protein interaction networks using xiSEARCH / xiFDR.
• Participate in integrative structural modelling (AlphaLink2) combining crosslinking MS with structural and bioinformatic data.
• Engage with research questions related to protein complexes, protein-drug interactions, and cellular mechanisms relevant to microbial physiology and antibiotic action.
• Work within an interdisciplinary environment at the interface of protein biochemistry, chemical biology, medicinal chemistry, analytical chemistry, and structural biology.

• Successfully completed scientific university degree (Master, Diplom, or equivalent) in Medicinal Chemistry, Pharmaceutical Sciences, Biochemistry, Biological Chemistry, Chemistry, or closely related discipline.
• Strong practical laboratory experience in at least one relevant area: protein biochemistry, molecular biology, analytical or physical chemistry, or chromatography.
• Solid understanding of instrumental analytical methods, ideally including chromatographic separation or spectrometric detection techniques.
• High motivation to learn and apply crosslinking MS, advanced LC-MS/MS workflows, and integrative structural biology approaches (is desirable).
• Strong interest in molecular mechanisms, protein structure, and interactions of proteins with small molecules or therapeutic agents, including antibiotics (is desirable).
• Ability to work independently with excellent organisational skills, accuracy, and scientific curiosity (is desirable).
• Very good communication and teamwork skills (is desirable).
• Excellent command of written and spoken English; German language skills are an advantage.
• Experience with X-ray crystallography or structural analysis tools (e.g., Chimera, PyMOL) (is desirable).
• Experience in peptide synthesis, protein sample preparation, enzymatic digestion, or peptide chromatography (is desirable).
• Background in medicinal chemistry, pharmacy, or related areas involving protein-targeted small molecules or mechanisms of drug action (is desirable).
• Familiarity with data analysis tools (e.g., Clustal Omega, sequence analysis, structural visualisation) (is desirable).
• Interest in interdisciplinary research bridging chemical biology, structural proteomics, and microbial systems (is desirable).
